چکیده

Follicular dendritic cells (FDCs) are unique immune system cells that contribute to the maintenance of humoral (i.e. antibody) immune responses. These cells are located in the follicles of secondary lymphoid tissues (e.g. lymph nodes and spleen), where they trap and retain antigens in the form of highly immunogenic immune complexes (ICs). FDC-ICs are composed of antigen plus specific antibody and/or complement proteins. These trapped antigens, combined with other soluble and membrane-bound signals contributed by FDCs, are essential to the development and maintenance of the germinal centre reaction and IgG and IgE responses respectively. FDCs also appear to contribute to initial IgM responses. In addition to their positive effects on humoral immunity, FDCsmay promote diseases includingHIV/AIDS (human immunodeficiency virus/acquired immunodeficiency syndrome), prion diseases and follicular lymphomas. A better understanding of FDCs should permit better regulation of antibody responses and may also allow the amelioration of some disease states.

تاثیر مایع رویی لنفوسیت T در القای بلوغ سلول‌های دندریتیک (DC1) و تولید سلول‌های موثر برای ایمونوتراپی تومور

Background: Nowadays, dendritic cells (DC) are used for tumor immunotherapy as they can induce immune responses against tumor cells. In this research, we comprehensively studied the maturation stimulus addition, PHA-activated T-cell (PHA- TCM) conditioned medium, autologous monocyte-conditioned medium (MCM) and TNF-α for their ability to promote uniformly mature dendritic cells that eli...

CD73 mediates adhesion of B cells to follicular dendritic cells.

Lymphocyte-vascular adhesion protein-2 was recently identified as CD73. The CD73 molecule, otherwise known as ecto-5'-nucleotidase, is a lymphocyte maturation marker that is involved in intracellular signaling, and lymphocyte proliferation and activation. We now show that CD73, in addition to mediating lymphocyte binding to endothelial cells, also mediates adhesion between B cells and follicula...

CD137 is expressed by follicular dendritic cells and costimulates B lymphocyte activation in germinal centers.

CD137, a member of the TNF receptor family, and its ligand are expressed on T lymphocytes and antigen-presenting cells (APC), respectively. During interaction with APC, T lymphocytes receive a potent, costimulatory signal through CD137. Reverse signaling has been demonstrated for the CD137 ligand, which causes activation in monocytes. Here we show that B lymphocytes also receive costimulatory s...

Follicular dendritic cells inhibit human B-lymphocyte proliferation.

In germinal centers, B lymphocytes are intimately associated with follicular dendritic cells (FDCs). It has been hypothesized that FDCs are involved in the regulation of B-cell growth and differentiation through cell-cell interactions. In this study, highly enriched preparations of FDCs were isolated by cell sorting using the FDC restricted monoclonal antibody DRC-1. When irradiated FDCs were c...

Adhesion of human B cells to follicular dendritic cells involves both the lymphocyte function-associated antigen 1/intercellular adhesion molecule 1 and very late antigen 4/vascular cell adhesion molecule 1 pathways

Presentation of antigen in the form of immune complexes to B lymphocytes by follicular dendritic cells (FDC) is considered to be a central step in the generation of memory B cells. During this process, which takes place in the microenvironment of the germinal center, B cells and FDC are in close physical contact. In the present study, we have explored the molecular basis of FDC-B cell interacti...
